Identification of the stem cell niche in the gastric mucosa of developing rats

Abstract

In gastric mucosa, the stem cells are located between the isthmus and neck of the gland, but in adult animals and imbalance situations, Troy cells can be activated and enter the cell cycle, generating a clutter which can lead to metalepsias . During development, the breastfeeding period is essential for the proper growth of the stomach, and the hormones and active milk peptides regulate the proliferation, differentiation and cell death. Our hypothesis is that if breastfeeding is an important regulator and development programmer, early weaning can modify the expression of genes that control stem cell niche and the cell cycle, and consequently altered growth, maturation and functioning of the gastric mucosa in adulthood. To test this hypothesis, this proposal has aims to identify the stem cell niche in the gastric glands of rats pups and evaluate the role of early weaning on these niches, specifically the distribution and location of Troy + cells in the gastric glands. During postnatal development, Wistar rats will be submitted to early weaning and the stomach will be collected at 18 and 60 days of postnatal life: detecting the stem cell marker Troy + by immunohistochemistry. The results will be grouped and analyzed statistically. (AU)
