Assessment of Mycoplasma hypneumoniae infection burden and immunopathology during the clinical phase of mycoplasmal pneumonia in experimentally infected swine

Abstract

The swine enzootic pneumonia (SEP) is a disease which causes high impacts in the worlwide swine producing industry, mainly due to the decrease of dialy weight gain. The causative agent of the SEP is the Mycoplasma hypneumoniae, a bacteria hard to culture and that has the pathogenic action in lungs not conpletely undestood. This research project focus on correlate and identify different features involved in the pathogeny an immunopathology of the disease wiht the severity and extension of lungs lesion in swine experimentally infected with Mycoplasma hyopneumoniae, strain 232. To this, 24 piglets are going to be used and 16 of them will be inoculated with 1,000,000 CCU/mL of the agent. The other 8 animals will be used as controls and are going to be inoculated with only 5mL of pure Friis medium. Periodically the animals will be submitted to clinical examination (cough index), blood sampling for antobody detection and BAL collection for qPCR analysis. At the 14th, 28th, 42nd and 56th days post infection, four infected animals plus two control animals are going to be euthanised and the lung lesions score assessed through differente coring techniques. Lung and trachea samples wil be collected and used for histopathology and cytokines gene expression of IL-1±, IL-1², IL-6, IL-8, IL-10, TNF-± e INF- ³. The data Normality is going to be detected using the Shapiro-Wilkins test. In case of Normal distribution is detected an ANOVA analysis will be performed altogheter with the simple T test and paired T test to compared results among groups and between different times. In case of the absence of normality in the data distribution non-parametric teste are going to be used (Wilcoxon test). In order to have a better assessment of the correlation between lung lesions, bacterial burden and the cytokine gene expression we will use the Pearson or spearman correlation test and regression analysis.
