Abstract

Periodontal disease (PD) has as a primary etiologic factor the oral biofilm. However, the disease results from complex interactions between the biofilm and the host immune response, which is responsible for up to 85% of tissue damage associated with periodontal disease. Within the new strategies of treatment host modulation was introduced in dentistry relatively recently. Interleukin 6 (IL-6) is a potent stimulator of osteoclast and bone reabsorption differentiation. Excess amounts of IL-6 are produced orally in patients with periodontitis. Tocilizumab, formerly known as myeloma receptor antibody (MRA), is a humanized monoclonal IL-6 receptor antibody that inhibits IL-6 / IL6r complex dimerization, avoiding signaling. Thus, taking into account advances in the pathogenesis of periodontal disease, its interactions with systemic diseases and new techniques for modulation of host inflammatory response, this study aims to evaluate the role of the IL6 receptor antibody in rats exposed to periodontal disease induced by ligature. 90 healthy Wistar Hannover SPF rats will be divided into 10 groups according to dosage and sacrifice time (n = 9): Control (C) -7 days, C-DP-7 days, C-DP-TC2-7 days, C- (C) -15 days, C-DP-15 days, C-DP-TC2-15 days, C-DP-TC4-15 days, C-DP-TC8-15 days. The outcomes will be evaluated through histometric, histochemical, micro-tomographic, gene expression (PCR array) and microbiome analyzes. (AU)
