Abstract

Cell communication, either in physiological or pathological conditions, is mediated mainly by molecules present in cellular surface as well as via soluble factors secreted by cells. Recently, a great interest has emerged regarding the mechanism of cell-to-cell communication through vesicles called exosomes. These structures are produced by many different cell types and can modulate cellular activity by induction of epigenetic alterations. Among the biological mechanisms modulated by exosomes, the tumor invasion process has been highlighted in recent studies. In this context, the present study aims to evaluate the in vitro effect of benign myoepithelial cell derived exosomes on the invasion of malignant neoplastic epithelial cells. For this purpose, we propose: 1) isolate and quantify exosomes derived from benign myoepithelial cells; 2) analyze a gene expression of metalloproteases 2 and 9 in malignant epithelial cell cultures exposed to the exosomes derived from benign myoepithelial cells; and 3) quantify the invasion of neoplastic malignant epithelial cells in Matrigel after exposure to exosome derived from benign myoepithelial cells.
