Abstract

The gastric body is the main region responsible for the digestive process, and has thick mucosa formed by tubular glands that open in pits on the surface. In adult individuals, these glands are divided into three segments: isthmus, colon and base and different epithelial populations are present. Stem cells are located between the isthmus and the neck region of the adult gland, but during postnatal development this niche is not yet determined, and proliferative cells are distributed throughout the gland. Despite the various studies and the great debate about the characterization of the gastric stem cell, it is not known which cell markers are present and the organization of the microenvironment, in terms of growth and signaling factors, is unknown. In addition, since the dietary pattern can alter the distribution of proliferative cells, it is necessary understand if and how the determinants of the stem-cell niche are influenced by diet. Thus, considering that breastfeeding plays an important role in the development of the gastric mucosa, early weaning (a common social practice) can lead to diverse changes, and also affect the gastric niche compartment. Our proposal aims is to evaluate the elements that form and establish the niches of stem cells and that determine their proliferation and migration, both in puppies and in adults, and to observe the immediate and late effects of early weaning in this population. During postnatal development, Wistar rats will be submitted to early weaning and the stomach will be collected at different ages for experiments in evaluation of gene expression, cell migration, detection and localization of molecules important for niche establishment. The obtained results will be grouped and analyzed statistically. (AU)
