Abstract

The main functions of the gastrointestinal tract are the digestion and absorption of nutrients mediated by specialized cells of the epithelial tissue. The activity of development, renewal and regeneration is controlled by the niche of intestinal stem cells located at the base of the crypts, however it is still unknown how this niche of proliferative cells behaves in face of alterations in dietetic pattern. Considering breastfeeding as an important role in development of intestinal mucosa, early weaning may promote changes in the process of cell differentiation and proliferation. Recent studies have analyzed epigenetic patterns of intestinal epithelial cells and demonstrated that the plasticity of the cell is maintained by a permissive epigenomic state and the specification of cell line is directed by activation of signaling pathways and expression of factors and transcription. Therefore, the aim of this proposal is evaluate markers related to the process of cell proliferation, differentiation and growth factors of the intestinal epithelium as well as possible epigenetic variation observing the immediate and late effects of early weaning. During postnatal development, Wistar rats will be submitted to early weaning and gut will be collected at different ages for evaluation of gene and protein expression, detection and localization of important molecules for the establishment of the niche. The obtained results will be grouped and analyzed statistically. (AU)
