Abstract Xanthomonas citri subsp. citri is the causal agent of citrus canker, an important disease to citrus farmers worldwide. This disease has no cure, and spreads rapidly by water droplets, penetrating in plants through wounds and, eventually, developing the canker itself: erumpent, corky lesions, which harbor bacterial growth on their surrounding areas. Attempts to break the disease cycle necessarily require bacterial inoculum to be eliminated - or at least diminished - at the orchards. Chemical control has limited efficacy and besides inducing the appearance of resistant strains it can also be a trigger for persister cells induction. Persisters are phenotypic variants that, due to low metabolic rates, are considered dormant. They present, among other characteristics, high tolerance to antimicrobial compounds and may be involved with recurrent cycles of disease. Here, we propose an in-depth study of persistence in X. citri, describing for the first time details for a plant-pathogenic bacterium. Besides, we aim to test and understand the ways these cells "wake up", which can be important for establishing future approaches for disease control and the re-designing of management practices to avoid the presence of live inoculum under field conditions.
