Evaluation of cytotoxicity and osteogenic potencial of experimental paste containing calcium hydroxide and activated charcoal as intracanal medication in endodontics

Abstract

Calcium hydroxide is the most commonly used intracanal medication in endodontics, and its purpose is to reduce the remaining microbiota to the chemical-mechanical preparation, control resorptive and inflammatory processes, and allow with the apical and periapical repair. Although it presents excellent characteristics, calcium hydroxide does not act effectively on biofilms of Enterococcus faecalis and Candida albicans, present in resistant endodontic infections. Thus, the association of this drug with other substances, such as activated charcoal, may amplify this antimicrobial effect. However, this association should not negatively interfere with the bio-compatibility and bioreduction of intracanal medication, because they are fundamental to the repair. Therefore, the objective of this study will be to evaluate the cytotoxicity of two experimental pastes containing calcium hydroxide and activated carbon, as well as to analyze the inductive potential on osteoblasts. For analysis of cytotoxicity, the 3- (4,5-dimethylthiazolyl-2-yl) -2,5-diphenyltetrazolium bromide (MTT) assay will be used in individual cultures of fibroblasts and osteoblasts. For the bioreductive potential analysis, the presence of alkaline phosphatase in cultures of osteoblasts will be measured. All experiments will make in triplicate and the data will be analyzed statistically by means of an appropriate statistical test, with a significance level of 5% (p <0.05). It is expected to determine the biological properties relative to cytotoxicity and the bioreduction of experimental pastes of calcium hydroxide and activated carbon.
