Development of toxicity test with sandollars Mellita quinquiesperforata eggs for ecotoxicological evaluation of chemotherapics tamoxifen, cisplatin and cyclophosphamide

Abstract

Nowadays exist several Contaminants of Emerging Concern, in other words, they are chemical substances which affect the environment, however, were not regulated yet. Among them, drugs can be cited, which are chemical substances biologically active used in treatment, control, and prevention of diseases. Along with the drugs, anticancer compounds stand out, whose mode of action can constitute an alkylating agent or changing the hormonal system. After the consumption, drugs are metabolized and eliminated in feces or urine, then being present in the sewage. The sewage treatment stations aren´t efficient to remove the Active Pharmaceutical Ingredients who came from the drug metabolization. This project consists to evaluate the toxicity of three anticancer drugs: Tamoxifen, Cisplatine, and Cyclophosphamide. It will be fulfilled toxicity tests of Mellita quinquiesperforata embryo development. The organisms will be collected in sandy beaches and stored in lab. The gametes collection will be done by osmotic shock (KCl 0,5N) or electric (9V). Afterward, fertilization in vitro will be done within the exam of newly fertilized eggs. For each substance, will be prepared 5 concentrations plus the control, being 4 replicates for each treatment, the replicates will present test-substance and about 500 eggs each. After 48h, embryos will be counted, aiming to evaluate development rates. For the data analysis (normal embryo development), it will be used ANOVA followed by Dunnett test. Therefore, it will be formed the firsts information about anticancer drug toxicity for Brazilian native marine species, assisting the sewage treatment management and marine biodiversity conservation.
