Effects of chronic central leptin infusion on food intake of offspring of rats with mild hyperglicemia

Abstract

Maternal diabetes is a metabolic disorder that confronts the fetus with an abundant supply of glucose, inducing adaptations that increase the susceptibility of offspring to developing diabetes and obesity in adulthood. Body weight is regulated by a complex system that includes peripheral and central factors. One of the hormones that plays an important role both in body weight regulation and in food intake is leptin, a hormone secreted by adipose tissue and an important signal that acts on the hypothalamus in regulating homeostasis of body weight and energy balance. Although several studies have shown the impact of maternal diabetes for offspring on a number of parameters, the effects on food intake control have not yet been fully elucidated. Thus, the aim of the present study is to evaluate the effect of chronic central infusion of leptin on food intake of offspring of rats with mild hyperglycemia. Sprague Dawley rats will be used. Experimental groups will be formed according to the presence or absence of maternal hyperglycemia and treatment with saline or leptin. Food intake and body weight will be monitored for 7 days before and 7 days after surgery for implantation of a cannula in the cerebral lateral ventricle. The cannula will be connected by a polyethylene tube to an osmotic pump (Alzet® model 2001) which will release saline solution or 3.5 ¼g / l leptin daily for 7 consecutive days. It is expected that chronic central infusion of leptin will reduce animal food intake and that this effect will be more pronounced in the offspring of rats with mild hyperglycemia. (AU)
