Evaluation of osteoinductive potential of a new rhBMP-2 in the reconstruction of large bone defects: a study in rats

Abstract

The aim of this study will be to evaluate the osteinductive potential of a new BMP2 in reconstructions after simulated bone resection in rats. For this, 36 male and adult rats (Rattus novergicus albinus Wistar), 6 months old, (n = 6 per group) will be subjected to bilateral femoral resection and reconstruction simulation, where stabilization will be performed by fixing means with 1.5 mm system titanium plates and screws and a 4 mm gap will be made. Thus, the reconstruction will be performed with rhBMP2 (Infuse) carried on collagen sponge (6.5 µg) and the three-dimensional framework maintained by conventional titanium mesh for the INFUSE Group, as well as the use of hBMP-2 (test) carried on collagen sponge (6.5 µg) for the reconstruction together with the titanium mesh for the hBMP-2 Group. For negative control group, the same resection, but reconstruction will not be performed with any interposition between the femoral stumps, and only the titanium - Mesh Group (negative control). The animals will be subjected to euthanasia by anesthetic overdose, and at 28 and 60 days After surgery for the evaluation of decalcified tissues, the femurs with the installed titanium plates, screws and mesh removed and fixed in formalin, will be descaled to EDTA (10%), for making slides for analysis histological analysis by staining Hematoxylin and Eosin, and analysis immunohistochemistry through proteins Runx2, Col-1, OPG, RANKL, OCN and BMP2. Within 60 days the parts prior to descaling will be analyzed through computed microtomography for evaluation of bone volumetric parameters (BV and BV / TV) and bone quality (Tb.Th, Tb.Sp and Tb.N). All quantitative data will be subjected to the normality curve. to establish the best statistical test (parametric or not) parametric), considering p <0.05 the level of significance.
