Evaluation of IL-10 cytokine expression after cellular exposure to AA extract in patients with periodontitis stage 4 grade c: in vitro study

Abstract

Although some studies show that pacients with Aggressive Periodontitis (AP) present specific differences in microbiological characteristics immunological and gene expression, its etiopathogenic factors are not yet fully established. Additionally, previous studies reported that AP has reduced levels of IL-10 in gingival crevicular fluid. However, the biological justifications for the occurrence of these differences in IL-10 production in both AP and Periodontal Health (PH) patients have not been fully investigated. Thus, the objective of the present study is to evaluate the protein expression profile of fibroblast cells isolated from individuals with AP and Periodontal Health (PH) and then to evaluate whether this protein expression profile is altered by bacterial stimulation by Aggregatibacter actimycetemcomitans (Aa), both for the AP group as for the PH group. For this, 9 individuals with AP and 9 individuals with PH will be selected and biopsies collected for the cultivation of gingival fibroblasts. After Cell Cultivation and expansion, cells from two groups will be stimulated with Aa and PMA + Ionomicin (PMA). After stimulation of the supernatant from each well, it will be specified as applied to inflammatory cytokines IL-1², IL-4, IL-6, IL-10, IFN-y, TNF-±. rom the results, it is possible to determine whether each stimulus (Aa / specific or PMA / nonspecific) affects cellular behavior, so that stimulation with A. actinomycetemcomitans could be determinant for a change in cytokine production in patients with or without AP. The results will be compared, after normality test, with the significance level of 5%.
