Evaluation of the antimicrobial and antibiofilm effect of the combination of calcium hydroxide and curcumin on Enterococcus faecalis monospecies biofilms

Abstract

Preservation of the deciduous tooth after irreversible inflammation or pulp necrosis has been considered a challenge for pediatric dentistry. In such cases, the modality of the chosen treatment is pulpectomy. In this sense, it is desirable that the material may contribute to the maintenance of pulp cavity decontamination, exhibiting antimicrobial properties. The aim of the present study will be to evaluate the possible antimicrobial action of calcium hydroxide pastes associated withcurcumin and different vehicles for filling primary teeth. The present study will be divided into two phases: I) planktonic phase - Enterococcus planktonic cells faecalis ATCC 29212 grown over 24 h will be subjected to the Minimuminhibitory concentration, maximum bactericidal concentration, metabolism assay byresazurin and checkerboard dilution, to test the antimicrobial effect of different concentrations of calcium hydroxide and curcumin solutions using propylene glycolas excipient; II) biofilm phase - E. faecalis biofilms will grow on specimens of bovine dentin and then will be treated with a) saline (control), b)calcium hydroxide + propylene glycol, c) curcumin + propylene glycol or d) curcumin +calcium hydroxide + polyethylene glycol. After 14 and 21 days of treatment, will be analyzed colony forming units (CFU) count and biofilm vitality intact by confocal microscopy. Statistical analysis will be conducted according to the normality and homogeneity of the data. P values <0.05 will be considered significant.
