Abstract Wound healing is conducted by sequential phases, and among the complex network of humoral and cellular events can be highlighted the activity of fibroblasts, which secrete cytokines and growth factors, actively participating in the development of new tissue and the regulation of the repair process. Several factors can compromise the mechanisms involved in the repair process, delaying and/or preventing its natural course, making the treatment of wounds to have limited success and including high-cost strategies. Several studies about the healing properties of natural products have been carried out because they can facilitate the healing process, since they demonstrate from anti-inflammatory, antioxidant, antibacterial actions to the synthesis of pro-collagen. Flavonoids are presented as strategic alternatives for various immunoinflammatory disorders, as they have a strong anti-inflammatory and antioxidant potential. The aim of this project is to evaluate the hypothesis that the flavonoids, eriocitrin (Erioc) and eriodictiol (Eriod), have modulating properties on the in vitro activity of L929 fibroblasts. L929 fibroblasts will be cultured with different treatments, according to each group - 1-Cells (Control), 2-Cells+ Erioc1µM, 3-Cells + Erioc25µM, 4- Cells + Erioc50µM, 5-Cells + Eriod1µM, 6-Cells + Eriod25 µM, 7-Cells + Eriod50 µM. The following analyzes will be carried out: cell proliferation and cytotoxicity (Alamar blue and MTT - 1, 3 and 7 days); migratory activity (scratch test - 24 and 72hrs); collagen synthesis (Hydroxyproline and Picrosirius - 3 and 7 days); expression of FGF and TGF-²1 growth factors (ELISA - 3 and 7 days). The data obtained will be analyzed statistically using the GraphPad Prism 6 software. (AU) | |
