Abstract

Organoids are structures formed in vitro that resemble organs in vivo, originated from pluripotent stem cells (PSC) or adult stem cells (ASC). Studies involving interaction of fungal pathogens on organoids are scarce. Therefore, there is great interest in deepening the understanding of the use of this technology to produce information from the infection, development and treatment of fungal infections. Gp43 is a 43kDa glycoprotein from Paracoccidioides brasiliensis and the P10 peptide, originated from a specific portion of the gp43, has been investigated as an experimental vaccine. Studies using encapsulated peptide P10 nanoparticles have shown that the use of nanocarriers assists in the effect of antifungal therapy against paracoccidioidomycosis. The main objective of this project is to use human 2D-lung organoid culture to elucidate the dynamics of fungal pathogen interaction and to evaluate the response obtained after contact of nanoparticles with encapsulated P10. Besides, standardization of the infection model in human 3D-lung organoid will be carried out, as well as the production and maintenance of the 3D culture cell. (AU)
