Effect of the association of different cryoprotectants on skin tissue slow freezing of Mazama bororo (Mammalia, Cervidae)

Abstract

The genetic diversity of the species can be preserved with the somatic cell cryopreservation, a good alternative for threatened species, as Mazama Bororo, this kind of cryopreservation allows to bank a lot of cells from an animal even after its death, that's an important source for cytogenetic studies and reproductive biotechnologies. The objective of this study is to test protocols for the cryopreservation of Mazama Bororo skin tissue fragments, using different cryoprotection by a slow freezing method. Five treatments will be evaluated, composed of three cryoprotectants that will be used alone and associated. The first one is the dimethylsulfoxide (DMSO), the second one is the ethylene glycol (EG), the third one is the dimethylsulfoxide (DMSO) associated with polyvinylpyrrolidone (PVP), and the fourth one is the EG with PVP. The last one is the control group that will not be cryopreserved. The skin tissue fragments will be collected from the medial region of the pelvic limbs. Each fragment will be divided into ten equal parts, patterned by weight, each animal will have two fragments for each treatment. The fragments will be collected and evaluated in cultivation, through the speed of the bottle occupation, into daily photos. Besides will be evaluated the mitotic index, to understand the toxic effect and the freezing effect into the capability of cell division. (AU)
