Evaluation of set and crystalline formation of a calcium silicate-based sealer in root canals dry and with moisture

Abstract

The setting is a needed factor for a root canal sealer to exercise its properties and dimensional stability in the root canal, however, this process is unsere for ready-to-use calcium silicate-based root canal sealer, which requires moisture to occur this reaction. Therefore, the aim of this study will be to evaluate the setting and crystalline formation of the ready-to-use bioceramic sealer, Bio-C Sealer, with the sealer in powder/liquid form, BioRoot RCS, in the cervical, middle and apical thirds of bovine teeth in dry and moisture root canals. The chemical evaluation of the setting of cement will be through Raman spectroscopy, FTIR, and X-ray diffraction (XRD). The superficial crystalline evaluation will be done through scanning electron microscopy (SEM) with energy dispersive spectroscopy (EDS). The sealer setting time will be evaluated using the ISO 6876/2012 standard. For this, 32 bovine teeth will be acquired. The teeth will be clean and will have the crowns cut from the root. The canals will be instrumented up to the #80 file and irrigated with 2.5% NaOCl for decontamination and removal of organic tissue. A Largo 6 will be used to widen the canal up to 2 mm in diameter. In the tests of setting time, Raman, and XRD, the same teeth will be used (n = 12) and distributed in each sealer group with dry (n=3) and moisture (n=3) subgroups. The functional chemical group's evaluation of sealers in their setting reaction will be through Raman and FTIR spectroscopy in 1, 14, and 28 days. In FTIR, each subgroup will have 3 teeth. In SEM/EDS, each subgroup (n = 2) will be evaluated on days 1 and 28. In XRD, the chemical compounds will be evaluated on days 1, 7, 14, and 28. For set marking, a 50 mg Gilmore needle will be applied in each third and the marking will be evaluated by 3 scores in 1, 7, 14, and 28 days. For Raman, FTIR, MEV/EDS, and XRD tests, the data will have a qualitative and semi-quantitative analysis. For a setting time, the data will be evaluated using mean and standard deviation, adopting a significance level of 5%.(AU)
