Abstract

Carbon dioxide (CO2) levels are increasing in the atmosphere and inside modern refrigerated buildings. High CO2 tensions (hypercapnia) are toxic to mammals by both pH-dependent and pH-independent mechanisms that remain partially understood. Relevantly, CO2 reacts with biologically ubiquitous oxygen metabolites such as peroxynitrite and hydrogen peroxide to produce carbonate radical and peroxymonocarbonate, respectively. The accumulated knowledge on these redox metabolites indicates the relevance of further investigations on their roles in cell physiology and pathophysiology. To this end, this project aims (I) to contribute to establish robust methods to detect peroxymonocarbonate in cells; (II) to examine whether the impairment of macrophages phagocytosis by CO2 depends on its redox metabolites; (III) perform global proteomic analysis of cells exposed to different CO2/HCO3- levels.
