Influence of dietary supplementation with lutein on the progression and treatment of experimental periodontitis induced in rats.

Abstract

The aim of this study will be to evaluate the effects of dietary supplementation with lutein (LT), both on the progression and as adjunctive therapy for the treatment of experimental periodontitis (EP). One-hundred-twenty male rats (Wistar) will be divided into two groups, no treatment (NT) or scaling and root planing (SRP). Then, will be subdivided into two subgroups receiving either sunflower oil (vehicle [VEH]) or LT (dissolved in VEH). EP will be induced in all groups by the placement of a cotton thread around the left mandibular first molar. Groups will be design as follows: NT-VEH, daily gastric gavage (GG) with 0.7ml/kg of VEH; NT-LT, GG with 250 mg/kg of LT; SRP-VEH, GG with 0.7ml/kg of VEH, and, 7 days after EP induction, ligatures will be removed and SRP performed; and SRP-LT, GG with 250 mg/kg of LT, and, 7 days after EP induction, ligatures will be removed and SRP performed. Euthanasia will be performed at 7, 15 and 30 days either after EP induction (NT-VEH and NT-LT) or periodontal treatment (SRP-VEH and SRP-LT). The collected hemimandibles with me submitted to histological processing for histometric analysis of the percentage of bone in the furcation (PBF), histopathological analysis of the periodontal tissues, and immunohistochemical analysis of TRAP, TNF-±, IL-1beta and IL-10. Data will be statistically analyzed (p d0.05).
