Abstract There is evidence that the increased incidence of differentiated thyroid cancer and thyroid diseases that has occurred in recent years is related to exposure to environmental factors, including endocrine disruptors (EDs). Glyphosate-based products such as Roundup® are widely used throughout the world and have been suspected of exerting ED effects and carcinogenic effects, but little is known about their mechanism of action. As abnormal cells may have different sensitivity, it is essential to determine the effects of this herbicide at different doses, both on normal and tumor thyroid cells. Our group has already been working with EDs (FAPESP nº 2020/02167-3) and we demonstrated that Roundup® has a proliferative effect on the Nthy-ori 3-1 lineage (of normal follicular cells) and, mainly, on TPC-1 (of papillary carcinoma) - Master's degree defended on 28/06, manuscript published in Frontier Endocrinology DOI: 10.3389/fendo.2022.904437. Continuing in this line of research, the objective of the present project is to evaluate other functional effects of Roundup® Original DI on normal and tumor thyroid cell lines exposed to different concentrations, including AOEL (Acceptable Occupational Exposure) and IDA (Acceptable Daily Intake) doses, according to ANVISA, such as genotoxicity, cell migration and invasion, in addition to analyzing the mRNA profile of these cell lines. We will use the following assays: comet, monolayer injury and transwell, and we will verify the genomic effects of the herbicide on mRNA expression by RNA-seq, next generation sequencing (NGS) in Nthy-ori 3-1 and TPC1 and BCPAP hyroid strains. | |
