Abstract

Introduction: The high consumption of industrialized diets contributes to the development of chronic metabolic diseases that are associated with early stages of several diseases, such as nonalcoholic fatty liver disease (NAFLD), a lifestyle- associated disease whose pathophysiological processes are contained in the metabolic syndrome. Molecular and Omics strategies are used in order to find targets for diagnosis and therapy based on detailed investigation of lipid composition, protein expression and the set of metabolites produced and/or modified in an organism. Gamma-oryzanol (³Oz) has shown protective effects in the treatment of metabolic disorders though its antioxidant and anti-inflammatory effect. The aim of this project is to investigate the Gamma-Oryzanol effect on microRNA expression, lipidomic, proteomic and metabolomic profile of the liver. Materials and methods: Animals will be divided into four experimental groups for 20 weeks: Control diet (C, n=8), C+³Oz (C+³Oz, n=8), high sugar-fat diet (HSF, n=8), and HSF+³Oz (n=8). The ³Oz will be added to diets to reach 0.5% of final concentration (w/w). Evaluation will include nutritional, metabolic and hormonal analysis, hepatic function, inflammation and oxidative stress. The microRNA-122 expression will be assessed through qPCR and the proteomic, lipidomic and metabolomic profiling of liver will be analyzed by label-free quantitative methods. Data will be presented by descriptive measures of position and variability and analyzed by specific comparison tests.
