Abstract

Human hemoglobin (Hb) inside red blood cells is responsible for oxygen (O_2) transportation from pulmonary alveoli to the tissues. However, when stored in blood bags (red blood cell concentrate) hemoglobins suffer loss of their functional properties and change in oxygen affinity, resulting in a decrease in the red blood cell concentrate quality used in blood transfusions. Thus, the objective of this work is to evaluate the effectiveness of hemoglobins stored in blood bags supplemented with the organic compounds 2,3-BPG (2,3-diphosphoglycerate), IHP (inositol hexaphosphate) and 5-HMF (5-hydroxymethylfurfural) through functional tests. Such compounds were selected from the literature because they have the ability to interact allosterically with hemoglobin, generating conformational changes in this protein and, possibly, producing an increase in its effectiveness in blood bags. This proposal will be linked to the Thematic Project Pathophysiological mechanisms and treatment of red blood cell abnormalities as part of the work (Fapesp Process 19/18886-1, Responsible Researcher: Prof. Dr. Fernando Ferreira Costa) as part of the Project "Structural-Function Correlation of Human Hemoglobin Variants" in which the guiding researcher acts as Associate Researcher.
