Histological analysis of paw pads to assess the effects of specific IL6R pressure on rates with ligature-induced periodontal disease and experimental rheumatoid arthritis

Abstract

Periodontal disease (PD), rheumatoid arthritis (RA) are immunoinflammatory diseases characterized by bone resorption induced by the inflammatory process (Mercado F, et al, 2000; Preshaw PM, 2017). Although these diseases have different etiologic factors, they all share similar risk factors and pathogenic mechanisms (Preshaw PM, 2017; Potempa J, et al 2017; Redman RS, et al 2016).Although the exact etiology of RA remains unknown so far, the production of pro-inflammatory cytokines and lymphocyte activation are crucial in disease events, just as it occurs in periodontal disease (de Aquino, S. G. et al., 2014; Lubberts, E. 2010; Lundy, S.K., et al., 2007).Thus, within the new therapeutic strategies, the modulation of the host's immune response seems to have great potential. Thus, taking into account the advances in the pathogenesis of periodontal disease, its interactions with systemic diseases and the new techniques for modulating the inflammatory response of the host, this study aims at the histological and histometric analysis in rats with induced RA and exposed to ligature-induced periodontal disease.32 Winstar Hannover rats will be used, divided into 4 groups (n=8): Group Naive (negative control), Group DP (periodontal disease induced by ligation), Group AR (rheumatoid arthritis induced by bovine collagen, Group AR/DP (RA induced by bovine collagen and PD).The animals will be euthanized on the 35th day.The results will be evaluated through histometric, histochemical, micro-tomography, gene expression (PCR array) and immunoenzymatic analysis.
