Analysis of keratin expression in keratinocytes exposed to photobiomodulation with simultaneous irradiation Utilizing 660nm and 808nm laser

Abstract

One of the direct effects of photobiomodulation (FBM) on the oral mucosa is the re-epithelialization in situations where tissue repair is stimulated. Recently, the duo simultaneous irradiation with 660nm and 808nm lasers has been adopted in the clinical practice of dentistry, but its mechanism of action is still poorly understood considering its effect on the cytoskeleton of keratinocytes. The objective of this study is to verify the effect of the simultaneous irradiation of the 660nm laser and 808nm on the expression of keratins in keratinocytes submitted to cell culture and wound assay. Immortalized keratinocytes (HaCat strain) will be maintained in cell culture, being divided into: Group I - without any treatment; Group II - irradiated with 660nm; Group III - irradiated with 808nm; Group IV - irradiated simultaneously with 660nm and 808nm. In the irradiated groups, will be adopted: 100mW, 20s of irradiation for the wavelengths isolated and 10s for simultaneous irradiation, 1.11W/cm 2 , 22.2J/cm 2. The cell viability test will be done using the MTS technique, and pair analysis of keratins 1/10 and 5/14 will be performed by immunofluorescence after the regular culture of keratinocytes and after wound assay culture. It is expected to detect differences between the groups regarding the pattern of cell migration and expression of keratins, contributing to the elucidation of the mechanism of action of simultaneous irradiation on the process of re-epithelialization and the differences in relation to irradiation with each wavelength separately.
