Aesthetic effectiveness and cytotoxicity of a manganese oxide-enriched at-home bleaching gel.

Abstract

The main objective of this study is to evaluate the effects of incorporating manganese oxide (MnO2) into a commercial carbamide peroxide (CP) gel on the esthetic efficacy and cytotoxicity of at-home bleaching therapy (HBT). Standardized enamel/dentin discs will be stained and then subjected to HBT for 15 days (2 hours/day), using a 22%CP containing or not MnO2, establishing the following groups: G1: No Treatment (Negative Control); G2: 22% CP Gel (Positive Control); G3: 22% CP Gel + 2 mg/mL MnO2; G4: 22% CP Gel + 6 mg/mL MnO2; G5: 22% CP Gel + 10 mg/mL MnO2. The total esthetic efficacy will be measured (n=8) through data collected after each bleaching procedure (”E00 and ”WI). For analysis of trans-amelodentinal cytotoxicity, the bleaching therapies will be performed on new enamel/dentin discs (n=8) adapted in artificial pulpal chambers. Then, the extracts (culture medium + components of the bleaching gels that diffused through the discs) will be obtained and applied to odontoblast-like MDPC-23 cells, which will be evaluated for viability (MTT assay). The amount of H2O2 diffused through the discs will also be determined (Leuco-crystal violet/peroxidase assay). The numerical data obtained will be subjected to specific statistical analysis.
