BIOPRINTING STRATEGIES FOR BONE REGENERATION BASED ON GELATIN METHACRYLATE BIOINKS FUNCTIONALIZED WITH METALLIC OXIDES

Abstract

This project aims the development of bioprinting strategies to obtain porous and bioactive scaffolds based on gelatin methacrylate (GelMA) associated to metallic oxides, personalized to bone defects. In phase 1, two bioprinting technologies will be tested to obtain GelMA scaffold containing a porous and interconnected internal microarchitecture. The effectiveness of these technologies to personalize bioprinted scaffolds on bone defect will be assessed by using 3D prototype models. In the second phase, GelMA hydrogels will be fabricated in association to distinct concentrations of magnesium, silicon and strontium oxide. An in vitro biological characterization will be performed to select potential formulations aimed at bone regeneration. In the last phase, after selecting GelMA formulations and bioprinting technique, the cell-homing potential will be evaluated using on-a-chip model, to evaluate pre-osteoblasts infiltration into scaffold structure and mineralized matrix deposition by migrating cells. At the end, the qualitative data will be analyzed descriptively and the quantitative data will be submitted to specific statistical analysis.
