Abstract

Preeclampsia (PE) is a gestational hypertensive disease characterized by hypertension and end-organ damage. It accounts for complicating 2-8% of pregnancies worldwide and impacts maternal-fetal health. PE can be classified into various subtypes, considering aspects such as onset of symptoms, severity degrees, and responsiveness to antihypertensive therapy - and some are associated with worse clinical outcomes. Metabolomics is the science of metabolites, which comprises compounds involved in several cellular processes and that may indicate phenotypic changes manifested by patients with PE. However, metabolome of pregnant women diagnosed with PE studies from the perspective of responsiveness to antihypertensive therapy is unprecedented. Thus, the objective of the proposed research project is to compare the circulating metabolomic profile in different subtypes of pregnant women with PE using bioinformatics tools with univariate and chemometric analyses. Subsequently, the effects of differential metabolites on human umbilical vein endothelial cell (HUVECs) lineage will be evaluated in terms of cell viability, oxidative stress, and endothelial dysfunction. Therefore, this study may contribute to understanding PE pathophysiology, investigation of future pharmacological targets and potential biomarkers for personalized treatments and better maternal-fetal prognosis.
