Histomorphic assessment of ligature-induced periodontitis in rats with experimental rheumatoid arthritis

Abstract

Periodontal disease (PD) and rheumatoid arthritis (RA) are immunoinflammatory diseases characterized by bone resorption induced by similar inflammatory mechanisms (Mercado F, et al, 2000; Lundy, S.K., et al., 2007; Lubberts, E. 2010 ; de Aquino, S. G. et al., 2014; Redman RS, et al 2016; Preshaw PM, 2017), having as one of the characteristics in common the greater expression of interleukin 6 (IL-6), a potent stimulator of the differentiation of osteoclasts and of bone resorption.The similarity between the pathologies points to a possible association when present simultaneously, suggesting a bidirectional relationship (Mercado et al., 2000; Lundberg et al., 2010, de Pablo; Dietrich; McAlindon, 2008; Chen et al., 2013; Mikuls et al. al., 2012). This correlation can possibly be expressed by comparatively observing the manifestation of diseases at bone levels.In this study, 32 Winstar Hannover rats will be used, divided into 4 groups (n = 8): Naive Group (negative control); Group C-DP (control with ligature-induced periodontal disease); C-AR group (control with bovine collagen-induced rheumatoid arthritis); RA/DP group (with bovine collagen-induced RA and PD). The animals will be euthanized on the 35th day.This proposal, part of the FAPESP Process 2021/02523-7 that studies the modulation of RA and PD with an IL-6 inhibitor, aims to evaluate, through histomorphometric evaluation, the influence of rheumatoid arthritis on periodontal disease, and vice versa, in rats with induced RA and exposed to ligation-induced PD.
