Abstract This research proposal aims to investigate the effect of applying orthodontic force on traumatized teeth of rats, after observation periods of 7 and 60 days, on the mechanisms involved in the process of dental repair after trauma. A model of tissue damage resulting from avulsion of upper incisor and treated with immediate replantation will be studied. Wistar rats will be randomly allocated into 8 groups (n = 24) according to trauma application, observation period, and implementation of orthodontic force. The animals will be euthanized after 1, 3, 7 and 14 days of force application. The following analyses will be performed: microtomographic analysis and morphometric analysis using optical microscopy to characterize resorption of the mineralized tissue; histopathological analysis with H&E; assessment of activity of clastic cells by TRAP histoenzymology; evaluation of the activity of MMPs in catabolism regions by in situ zymography; evaluation of the proteomic profile of the periodontal ligament by liquid chromatography-mass spectrometry; evaluation of protein expression patterns through bioinformatics analysis; and, in situ identification of differentially expressed proteins by means of immunohistochemistry. The data will be submitted to statistical analysis adopting a significance level of 5%. | |
