Abstract

The bioprinting of blocks for bone regeneration requires bioinks that combine great mechanical properties and high cell viability. The objectives of this study are to formulate bioactive bioinks for bone engineering, which presents high mechanical resistance, maintains high viability of periodontal ligament stem cells and promotes the osteodifferentiation of these stem cells. Two hydrogels from natural materials, namely gelatin, and chitosan of low viscosity will be synthesized in combination with hydroxyapatite modified by magnesium and strontium. Formulations will be adjusted to ensure that the printability of these hydrogels is optimized. Printability will be evaluated by extrusion capacity, filament quality and accuracy of the bioprinted construct in relation to the original design. Cell viability will be measured using the counting cell kit - 8 (CCK-8), 1, 7, 14 and 21 days after cell culture (n=4); Cellular differentiation will be measured by alizarin red assay at 21 days, in osteogenic and clonogenic medium. Compressive strength will be carried out on a universal testing machine (n=3). Data will be subjected to one or two-way ANOVA and Tukey's test (±=0.05).
