Abstract The root canal system is subject to invasion by microorganisms in various situations, such as deep caries or even iatrogenic events occurring during procedures. When not treated properly, these situations can initiate infectious processes due to the persistence of bacteria. This in vitro study aims to evaluate the anti-endotoxin action of medications N-acetylcysteine (NAC), calcium hydroxide [Ca(OH)2], and chlorhexidine (CHX), as well as their combinations, on the bacterium Fusobacterium nucleatum. The experimental groups of medications used are: SS (saline solution), NAC + Ca(OH)2, NAC + CHX, Ca(OH)2 + CHX, NAC + Ca(OH)2 + CHX. Following this, 50 morphologically and dimensionally similar teeth will be selected, instrumented with K#30 file, irrigated with 3ml of 1% NaOCl at each instrument change, and then filled with 17% ethylenediaminetetraacetic acid (EDTA) for 3 min, followed by a final irrigation of 10 ml saline solution. Subsequently, the specimens will be sealed apically with epoxy, and then the BHI broth containing the bacteria will be introduced into the specimens through contamination cycles. To assess bacterial penetration into the tubules, the specimens will be stained with propidium iodide and then analyzed under a confocal microscope. To evaluate the anti-endotoxin action, collections will be made at 3 different time points using sterile paper points and a chromogenic kinetic LAL assay, which will quantify the endotoxins. Statistical analysis will be performed using normality tests followed by analysis with Kruskal Wallis test complemented with Dunn's test or one-way ANOVA and Tukey's post hoc test. | |
