Abstract

Osteosarcoma (OS), is a primary bone tumor with prevalent occurrence in pediatric patients. Metastasis occurs in 80% of cases, primarily in the lungs, reducing survival to 20%. The process of metastasis in OS involves communication dynamics between OS cells and the tumor microenvironment, with exosomes released by these cells playing a central role. Exosomes encapsulate several molecules that can interfere with cell proliferation, immune system evasion, and metastasis. Studies highlight exosomal cargo's significance in OS development, particularly focusing on microRNA cargo. Collaborating with Dr. Tobias Bopp in Germany, a prominent expert in immunology, our study aims to analyze the contents of exosomes released by osteosarcoma cells and investigate the molecular targets involved in OS metastasis. Objectives include isolating exosomes, performing proteomic and secretome analyses, identifying differentially expressed molecules associated with metastasis, and validating potential targets in vitro and in an orthotopic mouse model. Anticipated outcomes involve discovering novel targets, contributing to a deeper understanding of OS metastasis, and paving the way for innovative treatment strategies to improve patient outcomes.
