Impacts of time-restricted feeding associated with physical exercise on obesity and skeletal muscle mass in an experimental model of shiftwork and high-fat diet

Abstract

There is evidence that a high-fat diet and sleep deprivation are important risk factors for increased body adiposity and sarcopenia. On the other hand, time-restricted feeding (TRF) and physical exercise have been strategies applied to combat these illnesses. However, it still needs to be explored whether TRA associated with aerobic and resistance physical training (combined training) may alleviate the health impacts of the experimental model of sleep deprivation (shiftwork) and high-fat diet. Thus, the objective of the present study will be to evaluate the effects of food restriction time alone or associated with combined physical training (CT) on obesity and skeletal muscle mass in mice subjected to sleep deprivation (shiftwork model) and fed a high-fat diet. Male C57BL/6 mice will be used, and distributed into different experimental groups, namely: control, fed with a standard diet; obese, fed a high-fat diet; obese subject to sleep deprivation (shiftwork); obese shiftwork subject to TRF; obese shiftwork subject to TRF associated with CT. The TRA will consist of 13 hours/day, carried out in the dark cycle. The CT will consist of aerobic and resistance exercises on alternate days, performed at the beginning of the dark cycle. After 8 weeks of intervention, assessments will be carried out: performance and strength, physiological (fasting blood glucose and glucose tolerance), histological (cross-section of skeletal muscle), and molecular (genes and protein content of the skeletal muscle hypertrophy pathway). The locomotor activity of the mice will also be analyzed. The findings of the present study will be of great value in understanding the impacts of TRA associated with combined physical training on obesity and muscle mass compared to an experimental model of shiftwork and feeding with a high-fat diet, allowing us to know whether there may be additive or complementary effects of these interventions.
