Local and systemic influence of resveratrol in hypertensive rats with apical periodontitis

Abstract

This project aims to analyze and compare the influence of resveratrol supplementation in hypertensive rats with apical periodontitis (AP) through local and systemic analysis of blood, heart and kidney tissues. Sixty-four 3-month-old Wistar rats will be used, 32 spontaneously hypertensive male Wistar albino rats (SHR) and 32 normotensives male Wistar rats. These animals will be distributed into 8 groups: normotensive (N), hypertensive (SHR), normotensive with apical periodontitis (N+AP), hypertensive with apical periodontitis (SHR+AP), normotensive under resveratrol supplementation (N+R), hypertensive under resveratrol supplementation (SHR+R), normotensive with apical periodontitis under resveratrol supplementation (N+AP+R), hypertensive with apical periodontitis under resveratrol supplementation (SHR+AP+R), with a sample number of 8 animals per group. The experimental protocol will last 45 days. On day 1, the initial blood pressure measurement will be performed and the daily supplementation with resveratrol (10mg/kg) by gavage will begin, which will continue throughout the experimental protocol. On the 15th day, the first molars will be opened, and the pulp will be exposed for 30 days to allow the development of the periapical lesion. On the 45th day, the final blood pressure will be measured, and the animals will be euthanized for blood collection, heart, kidneys and mandible. Throughout the experimental protocol, water and feed consumption, and body weight of the animals will be recorded. The blood collected will be used to characterize the blood profile through a complete blood count (cellular and biochemical parameters). In addition, blood plasma, cardiac and renal tissue will be used to analyze oxidative damage (COT, TAC, PC and TBARs) and antioxidant defense (CAT and SOD). Cardiac and renal tissue will also be used for histopathological analysis. The right mandible will be used for microtomographic analysis and the left mandible for histopathological and immunohistochemical analysis (IL-1¿, IL-10, TNF-¿, TRAP, OPG and RANKL). Statistical analysis will be performed using the SigmaPlot 14.0¿ program with a significance level of 5%.
