Abstract

INTRODUCTION: In liver transplantation, up to 10% of early liver failures are due to ischemia and reperfusion (IR). IR also increases the incidence of acute and chronic rejection, which may lead to re-transplantation. In addition, due to the current shortage of organs, extended criteria liver grafts have gained increasing acceptance for transplantation. In this context, strategies to minimize IR injury are needed. Previous research has shown that ischemic preconditioning (performing periods of IR of the organ for a short time) before prolonged ischemia reduces reperfusion-related injuries in normal mice. The aim of this study is to evaluate ischemic preconditioning in livers of obese (ob-ob) mice subjected to an ischemia and reperfusion model. METHODS: Twenty-four 8-week-old male ob-ob mice will be studied and divided into 3 groups: IR Group (N=8) - animals subjected to 15 minutes of hepatic ischemia and 1 hour and 30 minutes of reperfusion; PC+IR Group (N=8) - animals subjected to ischemic preconditioning (two episodes of ischemia followed by two episodes of reperfusion lasting 5 minutes) before prolonged ischemia of 15 minutes with reperfusion of 1 hour and 30 minutes; SHAM Group (N=5) - animals that will undergo the same previous surgical procedure but without hepatic ischemia. The biochemical parameters of liver function will be evaluated in an experimental model of hepatic IR in C57BL/6 ob/ob mice. (AU)
