Analysis of the osteoconductive potential of xenogenic bone substitutes functionalized with magnesium oxide by atomic laser deposition

Abstract

Faced with challenging situations in bone repair, it is of great interest to have alternatives that can optimize this complex process. In this sense, the use of biomaterials functionalized with magnesium oxide by atomic laser deposition is of great relevance. The aim of this study was to evaluate the effect of xenogenic osteoconductive bone substitutes based on hydroxyapatite, with different concentrations of magnesium oxides deposited by atomic laser action, on bone repair in wistar rats with critical size defects. To this end, 48 wistar rats will undergo surgery to make a critical size bone defect in the skullcap and then the defects will be filled according to the classification of the following sample groups: Blood clot (BC) without filling the defects; BIO-OSS with the defects filled with non-functionalized xenogenic bone substitutes; Mg200 with defects filled with functionalized bone substitutes with 200 cycles; and Mg500 with the defects filled with functionalized bone substitutes with 500 cycles. After 7 and 28 days post-surgery, the animals will be euthanized for sample collection and subsequent analysis, which will involve three-dimensional investigation by micro-computed tomography (Micro-Ct), histological analysis of the area of newly formed bone, inflammatory cell count and residual linear defect, as well as Picrosirius Red (PSR) analysis.
