Abstract

The regeneration of large bone losses is still a challenge in the medical-dental field. The advancement of guided bone regeneration is linked to the development of membranes that present high bioactivity with osteoinductive and osteogenic capacity. Different strategies can be associated to increase the bioactivity of such materials, such as incorporation of vitamin D and hydroxyapatite. The objectives of the present study are: to develop hybrid membranes of poly-L-lactide, vitamin D and hydroxyapatite and to evaluate the effect of such biomaterials on osteogenesis and osteoinduction of periodontal ligament stem cells (hPDLSC). Membranes will be made of pure PLLA or associated with vitamin D and/or hydroxyapatite, by means of co-solution electrospinning. Membranes will be characterized by scanning electron microscopy and contact angle measurement and vitamin D release will be evaluated by UV-Vis technique with absorbance at 260 nm. The proliferation and adhesion of hPDLSC will be measured using the CCK-8 kit at 1, 7, 14 and 21 days of culture. Cell differentiation in clonogenic medium and osteogenic differentiation medium will be analyzed by alizarin red assay after 21 days of culture. For each assay, respecting the requirements of normality and homoscedasticity, data will be submitted to single-factor ANOVA (mean fiber diameter and alizarin red) or two-factor ANOVA (cell proliferation), with an overall significance level of 95% (¿=0.05).
