Abstract The aim of this research is to get insight into microorganisms involved in autotrophic denitrification using sulfide as electron donor. For this, the stable-isotope probing technique (DNA-SIP) will be applied. Firstly, autotrophic denitrifier enrichment culture feed with bicarbonate, sulphide and nitrate will be obtained from a wastewater system operated in pilot scale. Denitrification will be followed in time by GC and HPLC methods. To identify the metabolically active microorganisms involved in this metabolism, actively growing autotrophic denitrifiers culture will be cultivated in medium with 13C-labelled bicarbonate, sodium sulfide and sodium nitrate. CsTFA separation of 13C-labelled DNA will be followed by PCR amplification DGGE analysis and sequencing of 16S DNA genes from microbial populations involved with 13C-bicarbonate metabolism. | |
