Abstract

Transplants therapies are normally limited by the organs donor quantity. One of the regenerative medicine goals is to find sources of cells and tissues for substitutive therapies. The embryonic stem cells play a promising source of transplant material by it exclusive propriety to be expanded indefinitely in culture (primary cells), being as a renovate source of tissue. In addiction, they are capable to differentiate in almost all cell types, being possible their utilization in substitutive therapies for many diseases. The challenge is to identify methods that promote it differentiation in specifics and functional adult lineages. Many studies have been isolate progenitors cells and demonstrated it abilities to proliferate for months and keeping it carotyps (Malhi, 2002), however the efficiency of differentiation is low. We propose the isolation of hepatic progenitor cells by your morphological and functional attributes from mice hepatic bud, focusing their isolation and culture to promote differentiation and posterior characterization of these cells in vitro for utilization in treatment of a mice lineage with hepatic insufficiency. This study also will characterize morphologically the hepatogenese process in mice, as the genes involved on cellular differentiation in this organ, furthermore we will create a hepatic stem cell and hepatic cell lineages cultivated in vitro bank for experimental studies.
