Abstract

Protoporphyrin IX (PpIX) is an intermediate in heme biosynthesis, synthesized within the mitochondria. As a photosensitizer in photodynamic therapy (PDT), PpIX primarily targets mitochondrial structures. This study investigates the interaction of PpIX with mitochondria isolated from mouse liver. Absorbance analysis reveals that exogenous PpIX added to mitochondrial samples in vitro binds significantly to the mitochondria. Fluorescence analysis of mitochondria isolated after delta-aminolevulinic acid (ALA) injection on mice demonstrates that PpIX accumulates within the mitochondria. This is a preliminary study for subsequent in vitro experiments, where PDT will be applied to mitochondria isolated from mouse liver, focusing on analyzing mitochondrial respiration. (AU)
