Evaluation of the tripliody in the susceptibility to the infection of Aeromonas jandaei and possible relation with the expression levels of immune related genes in lambaris (Astyanax mexicanus)

Abstract

Metazoans are mostly diploid, meaning that they have one pair of each type of chromosome in their somatic cells. However, some animal groups, such as fish, can present variations in the number of homologous chromosomes usually observed for the species, constituting a condition known as ploidy alteration. A type of alteration from the normal ploidy condition which is widely employed in fish production, as well as in research, is the triploidy, which is characterized by the occurrence of three copies of each chromosome in the genomic set. In most cases, this condition leads to reproductive sterility, which can be desirable in many situations, such as increase of body weight gain for the fish industry or for the production of generative chimeras for reproductive management of species that are difficult to reproduce in captivity. However, the biological consequences regarding disease susceptibility that triploidy may cause, and which immune parameters are affected by this condition, are still unknown. Thus, the aim of the present project is to characterize the sequences and analyze the gene expression of the cytokines TNF±, IFN³, IL-1², IL-8 e IL-10, the enzymes Arg2 and iNOS which are involved in macrophage activation, and the lysozymes LysC and LysG in response to the infection of a Brazilian strain of the bacterium Aeromonas jandaei in triploids and diploids specimens of lambari (Astyanax altiparanae), to evaluate the effects of triploidy on these fundamental parameters of the innate immune system of this commercially and environmentally important fish species.
