Abstract
The chelating agents Etidronate (HEDP) and Tetrasodium EDTA alkali (EDTANa4) recently were suggested as substitutes for EDTA in removing smear layer, therefore they possess the advantage they can be used mixed with sodium hypochlorite (NaOCl) in the biomechanical preparation of root canal system without compromising the dissolution of organic matter and the antimicrobial action of NaOCl. These substances may exert antimicrobial effect, if they are capable of chelating metals essential for microbial metabolism and stability of biofilm. The EDTANa4 is only available for use in research, but at the beginning of this year commercial form of HEDP is available in Europe, studies that evaluate biological effects of these rinsing become extremely relevant. This project will be useful to analyze the effects of mixtures of HEBP and EDTANa4 with NaOCl associated with ultrasonic agitation passive (PUI) on different aspects of the Endodontic treatment and to verify the effects of such as the contact with peripicais and rinsing in the demetalization of biofilms. Therefore, this project aims to: 1. Evaluate the effect adas HEBP blends and EDTANa4 with NaOCl associated with ultrasonic agitation passive (PUI) at debris removal of root canals and in extrusion of debris during instrumentation; 2. Evaluate the effect of HEDP, EDTANa4 and other rinsing on cell viability in NIH-3T3 cells culture and MC3T3-E1, in the capacity of mineralization, in the expression of pro-inflammatory cytokine genes by RT-PCR and the production of Pro-Inflammatory Cytokines by Western-blot MC3T3-E1 cells; 3. Evaluate the inflammatory response in rat subcutaneous by hematoxylin-eosin and Immunohistochemistry; 4. Develop a methodology to assess the order of biofilms by these chelating agents in confocal laser scanning microscopy. (AU)
