Abstract

Two non-inbred mouse lines, phenotypically selected for resistance (AIRmin) and susceptibility (AIRmax) to acute inflammatory response, show differential susceptibility/resistance to the development of several tumour types after chemical carcinogen treatment. An intercross pedigree of these mice has been generated and treated with chemical carcinogen dimethyl hydrazine, able to induce lung and intestinal tumours, at Instituto Butantan, Brazil. Thanks to collaboration with the Istituto Nazionale dei Tumori, Italy, the present project aims to carry out genome wide high-density genotyping with the innovative 2B-RAD technique to map genetic loci modulating multiple, phenotypes: individual genetic susceptibility to lung and intestinal cancer, and to acute inflammation. Moreover, it is planned to carry out a transcriptome analysis on lung tumour tissue and to map the expression quantitative traits loci (eQTL) that modulate the expression of genes related to the lung tumour phenotype. Overall, results of the project will provide an improved understanding of the relationship between genomic variation, individual genetic predisposition to tumourigenesis, and control of gene expression in tumours.
