Integrative transcriptomic and proteomic analyzes to identify biomarkers for feed efficiency, heat stress and tick resistance in Caracu breed

Abstract

Caracu is a taurine breed adapted to the tropical and subtropical Brazilian climate, maintaining the meat quality observed in taurine breeds combined with the ability to grow efficiently in any region of Brazil. The Caracu breed presents an excellent genetic material to help to elucidate the mechanisms that control complex traits. The project aims to identify biomarkers for heat stress, feed efficiency, and resistance to tick (Rhipicephalus (Boophilus) microplus) in Caracu cattle breed through integrative transcriptomic and proteomic analyzes. RNA samples will be obtained from peripheral blood, and the total RNA and miRNA libraries will be constructed and sequenced by HiSeq2500. The proteomic analyzes will be performed by gel electrophoresis (SDS-PAGE) and mass spectrometry (LC-MS / MS). The most significant genes will be validated by qPCR. The elucidation of the mechanisms that control heat stress and feed efficiency, as well as the understanding of the biological mechanisms responsible for vector-host interactions will contribute to the development of innovative strategies related to these traits. This research is a pioneer in report the integrative transcriptome and proteome of Caracu breed, from which genes, new alleles, functional RNAs, alternative transcripts, polymorphisms, proteins and new isoforms that contributes to the phenotypic differences observed for each trait will be identified.
