Abstract

The increased incidence of differentiated thyroid cancer and autoimmune thyroid diseases, Hashimoto's thyroiditis hypothyroidism in particular, may be related to exposure to environmental factors such as endocrine disruptors (ED). This project aims to investigate the cytotoxic and molecular effects of two ED: Bisphenol A (BPA) and glyphosate (Roundup Original®) on thyroid cells in vitro. BPA is an important component of many products present our daily lives, affects the binding of T3 to its receptor, acts as an indirect antagonist of thyroid hormones and appears to induce hypothyroidism, among other effects. Glyphosate is a widely used herbicide that appears to have a proliferating effect on thyroid cells. However, little is known about the mechanism of action of these ED in thyroid cells. We will use the CCK8 assay to quantify the viability of the cells tested; the Comet Test to identify DNA damage; the Micronucleus Test for cytogenetic damage and Next Generation Sequencing to identify possible genomic effects of these ED. Our research group has been working with cell culture for some time, and the current project involves an ongoing master thesis (Izabela Fernanda Dal' Bó Cruz - Grant CAPES). (AU)
