Evaluation of anti-inflammatory action of dipyrone and carprofen in a rat model of arthritis and dogs with cranial cruciate ligament rupture

Abstract

Dipyrone is a non-opioid analgesic widely used for its analgesic, antipyretic and spasmolytic effects. However, its mechanism of action has not yet been completely understood. Previous studies have shown that dipyrone produces an important inhibition of cyclooxygenases without causing adverse effects, which provides a great advantage over the use of non-steroidal anti-inflammatory drugs (NSAIDs). Experimental models are useful to understand mechanisms of diseases and drugs, as well as developing new drugs and providing diagnostic assistance. Several rat models of arthritis have been described, and a similar inflammatory process occurs in dogs with cranial cruciate ligament rupture (CrCL), reason why several studies have been using it as a model. This prospective, randomized, blind study aims to evaluate the anti-inflammatory effects, analgesia and side effects of dipyrone and carprofen in two different phases: experimental phase - subcutaneous administration of dipyrone at 177.8 mg/kg every 8 hours (Dr group), or carprofen at 5 mg/kg every 24 hours (Cr group), or tramadol at 17.8 mg/kg every 8 hours (group Tr) for 4 days in a rat knee monoarthritis model; and clinical phase - oral administration of dipyrone at 25 mg/kg every 6 hours (Dc group), or carprofen at 5 mg/kg every 24 hours (Cc group), or tramadol at 4 mg/kg every 8 hours (Tc group) for 4 days in dogs with cranial cruciate ligament rupture (CrCL). The modulation of inflammatory process and influence of prostaglandin E2 (PGE2) and the myeloperoxidase enzyme (MPO) activity will be investigated and computed tomography in rats and radiography, goniometry and thermometry in dogs will be performed.
