Clinical, hematological, biochemical and oxidative stress assessment of ozone therapy by rectal insufflation in healthy dogs

Abstract

Ozone therapy acts in the body causing a controlled oxidative stress, inducing an improvement in antioxidant, immune and circulatory responses, which seems to assist in infectious and inflammatory processes. However, very little is known about how ozone therapy affects the physiological parameters of animals, and further studies are needed to prove its safety. This type of evaluation is necessary to understand the pathophysiological mechanisms that ozone therapy causes in the body, an initial item so that this therapy can be recommended as a safe and effective treatment. In this context, the present study aims to evaluate the clinical, hematological, biochemical and oxidative stress parameters of healthy dogs submitted to ozonotherapy by rectal insufflation. For this purpose, at least 10 healthy dogs will undergo 4 ozone applications by rectal insufflation, at weekly intervals with collection of samples for laboratory evaluation twice a week. The hematological analysis will be carried out in an automated veterinary cell counter with differential leukocyte count and morphological analysis in a blood smear using optical microscopy. Biochemical analyzes will be performed in an automated photocolorimeter using a set of commercial reagents and oxidative stress will be determined by the total antioxidant capacity (TAC) by three methods: TAC equivalent to trolox by reducing the ABTS cation (TAC-ABTS), TAC by the reduction capacity plasma iron (TAC-FRAP) and copper-reducing antioxidant capacity (TAC-CUPRAC); in addition to the determination of total oxidative capacity (TOC), lipid peroxidation by substances reactive to thiobarbituric acid and the antioxidants uric acid and albumin. The variables will be tested for normality and the differences between the moments will be checked by ANOVA tests with repeated measures and post-test by Tukey or Friedmann with post-test by Dunn, being considered significant when p <0.05.
