Abstract

To better understand and try to elucidate the possible mechanisms of induction of genetic instability in professionals who have occupationally exposure to high levels of waste anesthetic gas (WAG) (results from FAPESP grant #2018/20900-0) and considering the lack of studies in the literature in professionals working in veterinary operating rooms (VOR), this original study aims to evaluate possible transcriptional, epigenetic, metabolomics and oxidative-inflammatory changes. Thus, professionals who are exposed to WAG isoflurane and sevoflurane working in VOR of a university hospital (exposed group) and individuals without exposure to WAG (control group) will be recruited. The expression of key genes related to DNA damage and repair/oxidative stress and inflammation; global methylation and expression of microRNAs (epigenetic assessments); plasma metabolomics; oxidative damage to DNA, lipids and proteins, antioxidant enzymes, antioxidant capacity and cholesterol and its fractions (oxidative stress) and inflammatory cytokines and high sensitivity C-reactive protein (inflammatory profile) will be evaluated. Epigenetic and metabolomics approaches are novel facing occupational anesthetic exposure and will certainly contribute to the study. WAG monitoring will be performed by both individual dosimeters and in the urine of professionals. Considering the importance and relevance of this topic, the results obtained will provide information that will contribute to a better understanding of the possible toxic effects/mechanisms of occupational veterinary exposure to WAG. Additionally, the project includes national and international collaborations, contributing to the consolidation of the internationalization of our research, as well as the training of human resources.
