Characterization of the inflammatory response in an experimental model of Extraparenchymal Neurocysticercosis

Abstract

Neurocysticercosis is the most common parasitic disease of the central nervous system. It occurs by the cerebral infestation by the larva of Taenia solium, which happens when eggs of the parasite are ingested. The prevalence of the disease decreased at the end of the 20th century due to sanitary measures, but it remains endemic in several regions of Latin America, sub-Saharan Africa and Southeast Asia. Migratory flows have determined the emergence of new cases in the United States and Europe. Experimental models of neurocysticercosis have been developed in the last decade, with a special focus on the parenchymal form of the disease. On the other hand, the extraparenchymal form of neurocysticercosis is more aggressive and raises more controversies, especially regarding therapeutic management, with lower levels of evidence for treatment recommendations. Therefore, we developed a model of extraparenchymal neurocysticercosis in rats by subarachnoid inoculation of Taenia crassiceps cysts. This model reproduces several radiological and morphological aspects of human neurocysticercosis and opens perspectives for laboratory investigation of the disease. In this project, we intend to investigate whether there is a change in the inflammatory pattern in extraparenchymal neurocysticercosis at different times of infection; whether sex is a determinant of morphological and inflammatory differences in experimental extraparenchymal neurocysticercosis; and to investigate the role of immunomodulatory drugs in the response to antiparasitic treatment. For this, we will employ histomorphometric analysis, enzyme immunoassay analysis for quantification of inflammatory cytokines and analysis of expression of genes involved in the inflammatory response through real-time polymerase chain reaction.
