Comparison of the anxiolytic effect of Valeriana officinalis, Diazepam and Nitrous Oxide during dental anesthesia

Abstract

The patient's anxiety is a common problem in dental procedures, especially during local anesthesia. In this respect, the aim of the present study will be to compare the clinical efficiency of different anxiolytic methods during local dental anesthesia, through analysis of the patient's hemodynamic parameters; measurement of salivary cortisol and pain, psychological and cognitive evaluation. For this purpose, the study will have randomized sessions, and each of the 25 participants will participate in two groups: G1 Diazepam 5mg and Placebo (O2), G2 Valeriana Officinalis 300 mg and Placebo (O2), G3 - Placebo and 50%/50% N2O/O2, G4 Placebo and Placebo (O2). Before any intervention, in both sessions, the participants will have vital signs measured, the trigger test and Corah's scale will be filled out, pre-procedure images will be presented, and basal salivary collection will be done. After these procedures, the participant will receive a capsule (test or placebo), and after waiting for 60 minutes, they will receive the gas mixture or only oxygen, according to the group they are assigned to. Then they will undergo the Gow-Gates anesthesia technique while vital signs are monitored, and saliva is collected. After the anesthetic procedure is completed, the participant will be asked to the pain/cognitive evaluation and amnesia tests. To investigate the effect of Diazepam and Valeriana Transient receptor potential vanilloid 1 we will performe a evaluation by calcium imaging experiments Fluo4AM-loaded. Data will be tested for normality and homoscedasticity and subjected to appropriate statistical tests. (AU)
